Flow Cytometry Protocol

Cell surface and intracellular staining for flow cytometry.

Cell Surface Staining

1. Harvest cells and wash with PBS + 2% FBS (staining buffer)
2. Count cells and resuspend at 1×10⁷ cells/mL
3. Add 100 μL cell suspension to each tube
4. Add fluorochrome-conjugated antibody (recommended dilution)
5. Incubate 30 min at 4°C in the dark
6. Wash 2× with staining buffer
7. Resuspend in 300 μL staining buffer
8. Analyze on flow cytometer within 1 hour
